Ophthalmology, along with other surgical fields, benefits from the unique properties of the amniotic membrane (AM), making it ideally suited for clinical applications. Its use is more widespread in situations requiring the repair of conjunctival and corneal damage. A retrospective study examined 68 patients with epibulbar conjunctival tumors, who received surgical treatment during the years 2011 through 2021. The surgical tumor removal procedure was immediately followed by AM application in seven (103%) patients. A breakdown of the cases revealed 54 (79%) malignant cases and 14 (21%) benign cases. Studies have shown that the NLRC3 gene, a member of the NLR family, containing a CARD domain, plays a key role in the intricate interplay of immunity, inflammation, and tumorigenesis. In spite of this, the clinical meaning of NLRC3 in the context of lung adenocarcinoma (LUAD) remains undefined. Publicly available RNA sequencing data and clinical outcome information were analyzed in this study to identify (i) NLRC3 as a tumor suppressor in lung adenocarcinoma (LUAD) and (ii) its potential in predicting patient responsiveness to immunotherapy. NLRC3 expression levels were found to be diminished in LUAD, exhibiting a more pronounced reduction in advanced-stage tumors. Moreover, a reduction in NLRC3 expression was observed to be associated with a poorer outcome for patients. NLRC3 protein levels exhibited a prognostic significance, which was also observed. Furthermore, a reduction in NLRC3 expression was observed, which inhibited the migration and infiltration of anti-tumor lymphocyte subsets and natural killer cells. Investigative mechanistic analysis indicated a possible association between NLRC3 and immune infiltration in LUAD, stemming from its role in modulating chemokine and receptor function. Still, the precise dosage control of DcEIL3-1 in the carnation petal aging process remains unresolved. Our screening of ethylene-responsive genes in the ethylene-induced carnation petal senescence transcriptome revealed two EBF (EIN3 Binding F-box) genes, DcEBF1 and DcEBF2, which demonstrated a substantial upregulation in response to ethylene treatment. Ethylene-induced carnation petal senescence was accelerated by silencing DcEBF1 and DcEBF2, but decelerated by overexpressing them, by affecting DcEIL3-1 downstream target genes but not DcEIL3-1 itself. Consequently, DcEBF1 and DcEBF2 cooperate with DcEIL3-1 to degrade DcEIL3-1 via an ubiquitination process, as observed in both in vitro and in vivo studies. To conclude, DcEIL3-1's binding to the promoter regions of DcEBF1 and DcEBF2 ultimately drives their expression. Our investigation into ethylene-induced carnation petal senescence has revealed a mutual regulatory relationship between DcEBF1/2 and DcEIL3-1. This finding not only deepens our insight into ethylene signaling pathways in carnation petal aging but also provides potential avenues for breeding long-lasting cut carnation varieties.
